【Purpose】
1. Master the operation method of protein sequence search;
2. Familiar with the analysis of basic properties of protein;
3. Familiar with protein function prediction based on sequence homology analysis, and understand protein function prediction based on motif, structure site and structure function domain database;
4. Understand protein structure prediction.

【Experiment content】
1. Use Entrez or SRS information query system to search human adiponectin (adiponectin) protein sequence;
2. Use BioEdit software to analyze the basic properties of the above protein sequences such as molecular mass, amino acid composition, and hydrophobicity;
3. Analyze the protein sequence of human adiponectin based on NCBI / Blast software;
4. Motif structural analysis of human adiponectin protein sequence;
5. Predict the secondary structure and three-dimensional structure of human adiponectin protein sequence.

【experimental method】
1. Search for human adiponectin protein sequence:
(1) Call the Internet browser and enter the Entrez website (http: // Entrez) in its address bar;
(2) Select protein in the selection bar after Search;
(3) Enter homo sapiens adiponectin in the input field;
(4) After clicking go, the serial acceptance number and serial name are displayed;
(5) Click the sequence acceptance number NP_004788 (adiponectin precursor; adipose most abundant gene transcript 1 [Homo sapiens]) to display the sequence details;
(6) Convert the sequence to FASTA format and save it (refer to the above steps to search the human adiponectin protein sequence using SRS information query system)

2. Use BioEdit software to analyze the basic properties of human adiponectin protein sequence such as molecular mass, amino acid composition and hydrophobicity:
Open the BioEdit software → enter the FASTA format sequence of the human adiponectin protein sequence into the analysis box → click the sequence description in the left sequence description box → click the sequence bar → select protein → click Amino Acid Composition → view the molecular weight and amino acid composition of the protein ;
Or after selecting protein, click Kyte & Doolittle Mean Hydrophobicity Profile → check the hydrophobicity level of the protein molecule;

3. Protein homology analysis of human adiponectin protein sequence:
(1) Enter the NCBI / Blast webpage;
(2) Select Protein-protein BLAST (blastp);
(3) Paste the FASTA format sequence into the input field;
(4) Click BLAST;
(5) View the homologous proteins;

4. Analysis of motif structure of human adiponectin protein sequence:
(1) Enter the http://hits.isb-sib.ch/cgi-bin/PFSCAN webpage;
(2) Paste the FASTA format sequence of human adiponectin protein sequence into the input field;
(3) Click Scan;
(4) View the analysis results (note the motif information in Prosite Profile);

5. Secondary structure prediction of human adiponectin protein sequence:
(1) Enter the website of the following protein structure prediction server:
http // (The PredictProtein Server);
(2) Click default in the You can column;
(3) Fill in the email address and serial name;
(4) Paste the FASTA format sequence of human adiponectin protein sequence into the input field and click Submit;
(5) View the analysis results from the email mailbox;

6. Three-dimensional structure prediction of human adiponectin protein sequence:
(1) Enter
http // SWISS-MODEL.html (SwissModel First Approach Mode) webpage;
(2) Fill in the email address, name and serial name;
(3) Paste the FASTA format sequence of human adiponectin protein sequence into the input field;
(4) Click Send Request;
(5) View the analysis results from the email mailbox (Note: you need to download the software into rasmol to view the three-dimensional image).
